Human studies using oral racemic mixtures of R- and S-ALA have observed plasma concentrations of R-ALA to be greater than that of S-ALA. One study found that, following oral administration of 50- or 600-mg racemic mixture of lipoic acid, maximum plasma concentrations of R-ALA were double that of S-ALA. Another study found tissue uptake of R-ALA was two- to seven-times higher than S-ALA.
A human study was conducted to assess the absorption and bioavailability of RLA bound to sodium (NaRLA), compared to both unbound RLA and a racemic mixture of R- and S-ALA. As anticipated from a previous study, NaRLA was shown to be more stable, more water soluble, and displayed significantly higher maximum concentrations (Cmax) and area under the curve (AUC) values – reflections of absorption – than unbound RLA or the racemic mixture.
